Hi Nicholas

Is it possible (I'm assuming using the .htaccess maker) to automatically block access to everyone to a certain file or directory?

A site of ours which was recently hacked had a folder added to the site which contained phishing content and although the folder is gone I can see there are a lot of requests coming through to it, I'm assuming from people looking at their emails and either the content being loaded there or then clicking on dodgy links within the email.

Can I use Admintools to just entirely block access to that folder and if so what is the best way of achieving it?

The easiest way would be to create this directory and put a .htaccess file in it with the following contents:
order deny, allow
deny from all

There are other more involving ways, creating custom .htaccess redirection rules, but I wouldn't want you to go there. A small mistake could block much more than you intended :)
