I'm usually using two profiles:

- daily onsite backup, count quota (2 to 7 depending on free space)

- daily offsite backup uploaded to S3 and deleted locally (important), maximum age 31 days, don't delete backups from 1st day of the month

So finally I need to make two backups, one after other.

Is it possible to configure one backup profile to do both things? I didn not find a solution so far, because count quota applies to local OR remote storage.

No, you cannot do that — and you don't actually need to. Having the backups on S3 means that you can pull them back to server either through Akeeba Backup (if you still have access to your site's wp-admin) or with Kickstart Professional (if you do not have access to your site's wp-admin).
